Ultra-High to Ultra-Low Drug-Loaded Micelles: Probing Host–Guest Interactions by Fluorescence Spectroscopy

The interactions between polymer micelle and encapsulated, hydrophobic drug are considered critical to control crucial formulation characteristics such as drug loading and drug release. Here, we probed the interactions between non-water soluble curcumin and two isomeric polymer micelles using spectroscopic methods. Unexpectedly, curcumin encapsulated in the high capacity micelle retained more flexible, whereas in the low capacity micelle, stronger hetero-intermolecular interactions occurred. Therefore, contrary to common perception, stronger interactions between host and guest can be detrimental to the drug loading in polymer micelles.
